Medvedev needed one hour and 22 minutes to confirm his 6-3 6-2 win, which tees up a clash with lucky loser Martin Landaluce, who beat Hamad Medjedovic earlier in the day.
But it was Tirante who made the faster start as he broke Medvedev's serve in the opening game of the match, only for the seventh seed to return the favour immediately after.
Medvedev then had to save a break point in a hard-fought fifth game before getting the better of Tirante's serve a game later, which helped him take the early advantage.
The second set started with a flurry from Medvedev, who opened with a break of serve, though the Russian squandered the chance to surge into a 3-0 lead soon after.
But Medvedev regained his composure and was on the brink of a place in the last eight, but against the serve, Tirante defended brilliantly to save three match points.
However, Tirante only delayed the inevitable as Medvedev ended a short rally with a deft shot at the net, which sealed his 23rd victory of the 2026 season so far.
